The FDA seizing Kratom

This came out of the FDA’s website most recent news on Kratom being attacked by the FDA at the american border.  This is bad news for Kratom advocates to see the FDA taking a harsh stance on Kratom leaves coming to america for human consumption.  What will it take for Kratom to be approved?  This plant must be available for research in order to give the necessary input to allow kratom to help people. Reason for Alert:
FDA has seen an increase in the number of shipments of dietary supplements and bulk dietary ingredients that are, or contain kratom, also known as Mitragyna speciosa, mitragynine extract, biak-biak, cratom, gratom, ithang, kakuam, katawn, kedemba, ketum, krathom, krton, mambog, madat, Maeng da leaf, nauclea, Nauclea speciosa, or thang. These shipments of kratom have come in a variety of forms, including capsules, whole leaves, processed leaves, leaf resins, leaf extracts, powdered leaves, and bulk liquids made of leaf extracts. Importers� websites have sometimes contained information about how their products are used.

Districts should be on alert to the possibility that the labeling, marketing, and/or promotion of kratom and kratom-containing products may indicate that such products appear to be unapproved and/or misbranded drugs. For such products, Import Alert 66-41 may be applicable.

Strains

You may notice when looking for kratom that there are too many types to keep up with. It would be prudent to consider that many names are fabricated for marketing purposes. If a source distingiush their kratom with a name, (other than Maeng Da, which translates in southern Thailand to a derogatory name for a pimp), it may be misleading if not proven to be unique by some intrinsic quality, hereditary or natural character, or trait. Professor Kratom trusts vendors that evidently have knowledge of where the trees that parent their kratom are growing, understanding of how this effects their harvested product, and have a deliberate method of preperation that ensures the preservation of quality and condition from the natural source, to our research facility. I encourage peers to scrutenize strains of kratom that claim “secret” origon and compare charicteristically with known strains.  

Two commonlaw ways strains are identified and often named are by location of the strain and the color of the vein bisecting the leaf.  Although this vein is often removed in most of the plant samples available, vein color is a genetic trait which alludes to the strains alkaloid contents.

Red-veined leaves have been recorded as having higher levels of  7-Hydroxymitragynine concentrations, but varied levels of Mitragynine, this pharmacologically translates to tranquility, euphoria, and other symptoms of mu opioid receptor aggregation, such as constipation, analgesia, constricted pupils. More Mitraphylline, known to be a vasodilator, muscle relaxant, diuretic, and boost the immune system, contributes to its profile as well.

I don’t think anyone describes White Vein kratom quite correctly. White Vein is my favorite – not because it gives a ‘rush of energy’, as people claim – but rather because it has a long come-up period that finally hits a peak at near 2 hours. And at the point I’m ‘glowing’ with happiness all over, but also very alert mentally.

I describe the White Vein feeling as being identical to that of tramadol, high dose. The onset & peak is similar too, as is the pronounced anti-depressant effect (euphoria). And I’ve long noticed that this quasi-tramadol effect is unique to White Vein strains only. Other kratom strains I’ve used just come & go too quickly, some even cause a slight fogginess. But the White Veins are very unique, thank god.

Kratom alkaloids are structurally similar to psychedelics however Kratom effects are more similar to mild Opiates. The effects of mitragynine are not reversed by the opiate antagonist nalorphine. The opiate-like effects of Kratom are caused by affinity to the delta and mu opioid receptors. low doses of Mitragynine tend to bind to delta receptors as well as alpha-adrenergic receptors. With higher doses, mitragynine will bind to mu receptors mimicking opioids whose effects are facilitated by the same action. Mitragynine and its derivatives actions in the central nervous system involve μ-opioid receptors, neuronal Ca²⁺ channels and descending monoaminergic projections resulting in central nervous system depression and local anesthesia. Acute side effects include increased urination, dry mouth, loss of appetite, and constipation. Large doses of kratom can cause overwhelming nausea and vomiting, but will normally subside after a lie down in the dark and a deep sleep. Too high of a dose could also produce a “hangover” depending on a persons metabolism and dose.

7-Hydroxymitragynine

7-Hydroxymitragynine is an active alkaloid in Kratom. It has a hydroxyl group at the C7 position of mitragynine, which was attributed the most potent effect, suggesting that the opioid effect of Mitragyna speciosa is mostly based on the activity of 7-hydroxymitragynine (Horie et al., 2005). It is fourfold the potency of mitragynine and has affinity to Kappa, Delta and Mu opioid receptors with higher affinity to Mu’s with a pKi value of 8.01+/- 0.02. More info on this and other Alkaloids coming soon!The potency, calculated using pD (2) values, was 30- and 17-fold higher than that of mitragynine and morphine, respectively. Antagonism of naloxone on concentration-response curves for 7-hydroxymitragynine confirmed its opioid effect. These results suggest that the opioid effect of M. speciosa is mostly based on the activity of 7-hydroxymitragynine.

Mitragynine

Mitragynine is the most abundant active alkaloid in the Kratom plant. In small doses its activity is reported to be stimulant like, while in higher doses more opiate like. These effects agree with the profile of mitragynine’s actions developed by study of its action on the brain. It has been shown to be adrenergic (like some related alkaloids from yohimbe) at lower doses while at higher doses mitragynine acts on the mu and delta opiate receptors. The mu opiate receptors are responsible for the enjoyable effects of the opiates, analgesia, and physical dependence. Its potential for treating drug addiction, perhaps in combination with ibogaine, is being investigated. It is orally active.

Mitragynine was isolated in 1907 by D. Hooper, a process repeated in 1921 by E. Field who gave the alkaloid its name. The alkaloid’s chemical structure was first illustrated completely in 1964 by D. Zacharias, R. Rosenstein and E. Jeffrey. It is structurally related to both the yohimbe alkaloids and, more distantly, voacangine. It is even more distantly related to other tryptamine-based psychedelic drugs such as psilocybin or LSD. Chemically, mitragynine is written 9-methoxy-corynantheidine.

Mitragynine itself acts primarily via μ-opioid receptors, although its oxidation product mitragynine-pseudoindoxyl, which is likely to be a major component of kratom that has been aged or stored for extended periods, acts as a fairly selective δ-opioid agonist with little affinity for μ or κ receptors. Another alkaloid with a major contribution to the μ-opioid activity of the kratom plant is the related compound 7-hydroxymitragynine (above), which while present in the plant in much smaller quantities than mitragynine, is a much more potent μ-opioid agonist.

Also it is STRONGLY recommended that you do not use kratom in combination with MAO inhibitors, such as Syrian Rue (Peganum harmala), Banisteriopsis caapi, Passionflower (Passiflora incarnata) and certain anti-depressants. Serious, even fatal, reactions can occur if MAO inhibitor drugs are combined with monoamine drugs. The combination of MAO inhibitor drugs with kratom, which contains monoamine alkaloids, has not been studied.
